You could buy spacer beads (glass, sapphire, ...) of given diameter and tolerance and use those, e.g. in combination with a UV curable adhesive that you wick in to permanently fix things. Also, the spacer needs to be fixed manually on the glass plate: it cannot be processed on the glass plate but it has to be attached afterwards.
